Aside from the physical positioning of these wolves (one on top, the other underneath), a good way to establish which wolf is an alpha is to look at the tail position. A lowered or tucked tail displays a beta (subordinate) or omega wolf. Typically when the tail is either straight out or up in the air, you have an alpha wolf. This and intended eye contact are the best ways to discover the alpha in a pack. (And also a way to avoid making the false assumption that the largest wolf is the alpha)
However, one could easily argue that this picture shows a challenge between the alpha and a beta. The alpha (holding the raised tail) is looking away from the beta (black) wolf. The beta seems to be staring straight into the alpha’s eyes. This reveals a possible battle for the position of leader occuring.
